Detection of metabolites of flavonoids Epimedium in rats urine by high performance liquid chromatography-high-resolution tandem mass spectrometry

A method for the detection of Epimedium flavonoid metabolites in the rat urine was developed on the basis of liquid chromatography co0mbined with high-resolution tandem mass spectrometry. The urine sample preparation procedure included a solid-phase extraction, evaporation of the organic extract in a stream of nitrogen and reconstitution of residue in mobile phase. Separation of urine metabolites was carried out by reversed-phase chromatography. For the data dependent acquisition mode, only product ions with a pre-set values of neutral loss were used. Six metabolites were detected and their structures were proposed on the basis of fragment ions characteristic of Epimedium flavonoids and fragmentation patterns of the detected compounds. Main metabolic reactions and proposed structure of the detected compounds were modeled.
